Sequence Analysis:
The Core Guarantee
Control reactions are run with every sequencing gel. Thus:
- There is no charge if the control reactions fail.
- There is no charge if we generated the template (Guaranteed
Results).
- Otherwise, there is a 50% charge for each failed reaction.
Primer
- We provide a free standard primer with a free charge. Please see the list on the sequencing order form.
- Standard Primer:
- M13F (-21), M13F (-47), M13R (-22),
- T7P Terminator, T7P Promoter
- SP6, T3
- KS and KS
- Non-standard primers should be carefully quantified spectrophotometrically (preferably scanned 230-280 nm) and provided with the concentration 20ng/ul. Submit 10ul for each reaction.
Primer Design
A few "rules of thumb" for sequencing primers
- 18-25 bases in length
- 40-60% G+C, preferably with 3' G or C
- No homopolymeric tracks of more than 3 bases
- Minimize secondary and tertiary structure
